Sperm DNA fragmentation doesn't happen in isolation; it's downstream of specific molecular failures during spermatogenesis. Recent genetics research shows that aberrant histone acetylation during sperm chromatin compaction is directly linked to increased DNA fragmentation. At the same time, imbalanced protamine-1 to protamine-2 ratios (which govern how tightly the genome is packaged) are separately tied to poor motility and fertility outcomes (1).
Those findings inspired our team to research predictive models built on sperm DNA methylation profiles, which have already shown high sensitivity in classifying male fertility status, which is a proof that molecular and epigenetic signals, not just morphology or motility, can meaningfully predict outcomes.
We're now working on incorporating these functional and epigenetic markers into our platform, extending our imaging-based analysis toward a more complete picture of sperm quality. More to come as this feature takes shape.
